Is there a theme running from something that happened during Asimov's personal life that carried over into his professional writing?

Absolutely.  Almost all of Forward to Foundation is somewhat autobiographical if you look at Seldon as Asimov.  First, the his advisers (editors) leave him, he is unwillingly thrust into a position of leadership, (sci-fi) his wife dies, (divorced) he finds a new woman to share his life with, (new wife) and dies with his life's work completed (works).
